You really shouldn't post your public IP-address, port number, username and password all together like this. Or do you want other people to try to connect to it?

Are you even sure that the API is accessible on the public side of that router? Having the API accessible to the public internet is a really bad idea. At the very least have a IP-whitelist on it.

Are you sure that error message isn't just because the router didn't reply or something?

Have you ensured that you can access it from your IP-address? Does the user have API permission?

Which RouterOS version are you using?

Avoid posting the public IP Address of your as possible for security. First try to access your mikrotik using a software called winbox and check if it is accessible. if accessible, also check if the API service is enable and on the same port by navigating to IP>Services.
